I was going to say something along the lines of scientists who discover an virus or bacteria that can't be eliminated but keeps spreading and changing people. But that has already been done several times over.
How about a few people from around the world get together via computers to play impossible trivia tournaments and ignore work, family and private lives in order to win 5 million dollars? This idea can be extended ......

Neon Genesis: Evangelion
It's an anime series that I have seen, and personally like, that I think would make a spectacular film....or film franchise.
The storyline would have to be altered for a more global acceptance, but 40 story tall, athletic cybernetic beings doing battle with mysterious Earth attacking organisms?
Dunno but anybody else, but the visions are swimming in my head.
